Course content
Lectures: 1. Basic characteristics of the immune system and its functions. Immune system as 3rd homeostatic regulatory system, structure, basic components, Characteristics of immune response - main principles and mechanisms of immune reactions. 2. Cells and molecules. Morphological and functional characteristics of immune system cells, surface molecules, adhesins. 3. Mechanisms of natural immunity. Phagocytosis, natural cytotoxicity; effects of interferons, acute phase proteins. 4. Complement and inflammation. Activation pathways, biological functions; inflammation - its importance in the system of defense reactions of the organism, cells and mediators of inflammation, time dynamics. 5. Acquired (adaptive) immunity. Lymphocytes - their differentiation, specific receptors for antigen, characteristics of individual subpopulations and their function in immune reactions; genetic basis of antigen binding site variability, antibodies - structure and isotype characteristics, primary and secondary antibody response, antibody effector activities. 6.-7. Main histocompatibility system and its immunological significance. Gene organization, structure, function. Antigen presentation and T cell activation; regulatory role of Th cells, Th1 x Th2 type of reaction. 8.-9. Cytokines and other regulatory mechanisms of immunity. Structure and function of immune mediators and their mutual relations, anticytokine therapy. Regulatory T lymphocytes. 10.-11. Immunohematology. Immunohematology and immunogenetics, definitions, basic terms. Blood cell antigens, blood groups: systems, collection, series - antibodies. Immunohematology of leukocytes and platelets. Examination methods in immunohematology. 12-13. Immunopathological reactions and importance for clinical immunology. Basic types and mechanisms of immunopathological reactions, general characteristics of immunodeficiencies, allergies and autoimmune diseases. 14. Nanotoxicology. Nanoparticle toxicity, relation to immunotoxicology, modern methods for study and characterization of nanoparticles. Exercises: Laboratory exercises are focused on practical exercises and verification of students' knowledge gained during lectures and study of recommended literature. CV1 Serology - agglutination, precipitation, complement binding, ELISA, WB, CLIA Principles of work in immunological laboratory, principles of serological reactions (pipetting, titration), basic principles of the given serological methods. CV2 Immunofluorescence, IGRA tests and diagnostics in allergology Principle and use of immunofluorescence in diagnostics of autoimmune diseases, principle of IGRA tests, basic approaches to diagnostics in allergology (sIgE, BAT). CV3 Fundamentals of flow cytometry, immunonephelometry Principles of flow cytometry, importance of monoclonal antibodies, immunophenotyping, lymphocyte subpopulation, respiratory burst test, principle of immunonephelometry and determination of serum protein levels (immunoglobulins, complement components). CV 4 Examination methods in immunohematology Principles and application of immunohematology methods. Examination of AB0 and RhD system, agglutinins and agglutinogens. Antibody detection tests, direct and indirect antiglobulin test, cold and heat antibodies. Importance for clinical practice.

Learning outcomes
Learning outcomes of the course unit The course provides students with basic information concerning human immunology. The lectures and practical laboratory exercises will discuss the basic structures and functions of the immune system, its importance in the defense against infectious and cancerous diseases, causes and mechanisms in the development of immune system diseases. In lectures are discussed mechanisms of non-specific and specific immunity, defense mechanisms in infectious and non-infectious processes and causes and mechanisms of immunopathological conditions. In practical exercises, the basics of serology and laboratory immunology are discussed.

Credit: Laboratory exercise 100% attendance, elaboration of protocols. Combined exam, oral and written part.
