Tato bakalářská práce se zabývá studiem degradačního chování PCL nanovlákenné vrstvy s inkorporovaným alaptidem. Pro srovnání byla pozorována zvlákněná vrstva z čistého PCL, která byla připravená za stejných podmínek. V teoretické části jsou popsány důvody, proč jsou pro tkáňové inženýrství využívány nanovlákenné materiály, spolu s technologií jejich přípravy. Dále jsou stručně charakterizovány možné biodegradabilní polymery, funkcionalizace nanovlákeného materiálu PCL pomocí metody zvlákňování z blendu, alaptid jako biologicky aktivní látka a mechanismus degradace nanovláken. Experimentální část je zaměřena na optimalizaci enzymaticky katalyzované degradace. Enzymem použitým při experimentu byla lipáza. Také je zde popsána metodika práce.áNejprve byla sledována morfologie připravených materiálů PCL. Poté v rámci degradačního chování PCL nanovlákenných vrstev byl sledován hmotnostní úbytek materiálu, změna morfologie materiálu a změna molekulové hmotnosti PCL během degradace. Výsledky ukazují, že inkorporovaný alaptid nemá vliv na rychlost degradace polymeru, ale má vliv na změnu molekulové hmotnosti polymeru.

This bachelor thesis focuses on the study of degradation behaviour of alaptide loaded PCL nanofibrous membrane. As the reference material, pure PCL nanofibrous membrane was prepared at the same conditions. Theoretical part of the thesis describes why nanofibrous scaffolds are used for tissue engineering, together with the technology of their design. In addition, description of biodegradable polymers often used in tissue engineering, functionalization of PCL nanofibres using blending electrospinning method, alaptide as a biologicaly active compound and the degradation mechanism of nanofibers are briefly described. Experimental part is focused on the optimization of enzymecatalyzed degradation. The lipase from Pseudomonas cepacia was used to catalyse hydrolytic degradation of PCL. The methodology of the work is also described. At first, the morphology of electrospun PCL membranes was analyzed. Then the degradation behaviour of PCL nanofibrous membranes was analyzed through the weight loss of the material, the change of the material morphology and the change of the molecular weight of PCL during degradation. The results show that the incorporated alaptide does not affect the rate of polymer degradation, only a change in the molecular weight of the polymer during degradation was detected in the material without alaptide.
